This dataset was used for solving an X‑ray crystal structure of hepatitis C virus (HCV) RNA‑dependent RNA polymerase, NS5B, from genotype 2a (strain JFH‑1) containing the substitutions S15G, E86Q, E87Q, C223H, and V321I. The structure was determined in complex with flavin adenine dinucleotide (FAD), cytidine diphosphate (CDP), Mn²⁺ ions, and an RNA template with the sequence 5′‑UGU. The model provides detailed structural information on FAD binding in the NS5B de novo initiation complex.
