Single-cell sequencing of human lacrimal gland tissue and organoids

Human lacrimal gland tissue and organoids, cultured in expansion or differentiation, were FAC sorted into plates to follow the SORTseq protocol (Muraro et al., 2016). Overall design: Human lacrimal gland tissue was obtained from the UMC Utrecht. Tissue was dissociated into single-cell and stained for EpCAM, NGFR or nothing. Then cells were sorted into plates. For human organoids, cells were cultured in expansion medium for 7 or 9 days and, to differentiate them, they were first split into expansion medium for 2 days then switched to differentiation medium for 7 days, as described before. Then, organoids were dissociated and FAC sorted.
