This study characterizes the development of an anaerobic intestinal cell culture model based on a dual flow chamber system using Caco-2 cells. One goal of the study was to characterize the transcriptomic profile of Caco-2 cells grown in the anaerobic dual flow chamber system and characterizing the effect of oxygen- and flow conditions on the maturation of the cells. Overall design: Caco-2 cells were cultured in the apical channel of a dual-flow chamber system, with or without deoxygenated medium (<1% O2 saturation), starting on day 7 post-seeding (DPS). These conditions were compared with Caco-2 cells cultured statically in Transwell inserts. RNA was harvested from the cells at 13 DPS.
