The aim of this study was to identify non-coding RNA that were differentially expressed 7 days after induction of osteogenic or chondrogenic differentiation in human bone marrow-mesenchymal stromal cells (MSCs). Overall design: Human MSCs were induced to osteogenic differentiation in monolayer (DMEM-LG, 10%FBS, 1%P/S, 50 µg/mL ascorbic acid 2-phosphate, 5 mM Ã-glycerol phosphate and 10 nM dexamethasone). Chondrogenic differentiation was induced in pellet culture using DMEM-HG, 1%P/S, 1% ITS+, 1% NEAA, 50 µg/mL ascorbic acid 2-phosphate, 100 nM dexamethasone, and 10 ng/ml TGF-Ã1. Media were refreshed every second day. At day 7, samples were collected in triplicate for each donor (n=3 donors) for RNA extraction. 2 samples for each condition and each donor were selected for RNAseq analysis.
