Non-canonical STAT3 activity sustains pathogenic Th17 proliferation and cytokine response to antigen Th17 WT (5 mice total, combined from two separate experiments) and Th17ΔSTAT3 (4 mice total combined from two separate experiments) were immunized with MOG(35-55) in CFA to induce EAE. On day 10 post-immunization dLN were harvested and CD4+YFP+ Th17 cells were sorted on a FACSAria II, directly into SmartSeq low-input RNA kit lysis buffer. Libraries were prepared using Nextera XT DNA library prep kits, RNA-Sequencing was performed on Illumina NextSeq500 by the Health Sciences Sequencing Core at University of Pittsburgh.
