This study aims to measure the transcriptome profile of ilium tissues from intestinal epithelial conditional Ifnar1 KO and control littermates following cryptosporidial infection. Neonates of the conditional intestinal epithelial Ifnar1 knockout mice and control littlermates were exposed to C. parvum infection for 48 and 72h. Ilium tissues were collected and total RNA was isolated for the genome-wide analysis. The conditional intestinal epithelial type I IFN receptor Ifnar1 knockout mice (Villin.Ifnar1-/- mice) were generated from cross-breeding of the Ifnar1fl/fl mice [B6(Cg)-Ifnar1tm1.1Ees/J, Jackson] with the Villin.Cre transgenic [B6.Cg-Tg(Vil1-cre)1000Gum/J, Jackson]. Neonates of Villin.Ifnar1-/- mice and littlemates (5 days after birth) were administrated with C. parvum oocysts through oral administration. Neonates received oral adminstration of PBS were used as control. At 48h and 72h after parasite (or PBS) adminstration, ilium tissues were collected and total RNAs were isolated with the RNeasy Mini kit (Qiagen) according to the manufacturer’s instruction. High-throughput sequencing service was provided by the BGI Americas.
