Gene expression profiles of ASC52 cells differentiated to mature adipocytes and dedifferentiated by MDA-MB-231 cells

It is widely recognized that cancer development and progression depend not only on tumor-cell intrinsic factors but also on its microenvironment and on the host characteristics. Adipocytes are the main stromal cells in the breast and an heterotypic interaction between breast epithelial cells and adipocytes has been demonstrated. To date, the alterations associated with adipocyte dedifferentiation has to be further studied. The aim of our work is to compare gene expression profile of mature adipocytes (MA), adipocytes dedifferentiated (DED) by breast cancer cells and mesenchymal stem cells (MSC) using the ASC52htert in vitro model. ASC52htert adipocyte derived mesenchymal stem cells (ATCC) were seeded in MesenPRO RS (GIBCO) and 3 days after differentiated or not to mature adipocytes culturing them in the StemPRO Adipogenesis Differentiation medium (GIBCO) for 14 days. Dedifferentiation was induced by culturing ASC52 mature adipocytes for 7 days in conditioned media of MDA-MB-231 cells (RPMI, 10% FBS). Mature adipocytes after differentiation were cultured for 7 days in control medium (RPMI,10% FBS). Cells were harvested, washed in PBS 1x and immediately stored at -80°C until RNA extraction.
