RNA-seq study of transcriptiona factor FleQ in Pseudomonas putida KT2442. Pseudomonas putida

We constructed fleQ mutant strain and analyzed its regulation role through gene expression profile compared to the wild-type KT2442. Overall design: The Pseudomonas putida KT2442 in-frame deletion fleQ mutant and wild-type KT2442 were analyzed by RNA-seq. All samples were cultivated in LB medium until reaching an OD600 of 1.3 or 3.5. Cultures were pooled for RNA extraction, library preparation and sequencing. qRT–PCR validation was performed using SYBR Green assays.
