This project characterizes transcriptomic responses in human renal proximal tubular epithelial cells (HK-2; Homo sapiens) after exposure to the PFOS substitute F-53B (chlorinated polyfluorinated ether sulfonate). HK-2 cells were cultured in DMEM/F12 with 10 percent heat-inactivated FBS and 1 percent penicillin-streptomycin at 37 C with 5 percent CO2, then treated with F-53B or vehicle control for 48 hours. Total RNA was isolated using TRIzol, and RNA quality control, library preparation, and sequencing were performed by Beijing Qingke Biotechnology Co., Ltd. Raw FASTQ reads from control and F-53B treated samples are deposited to enable differential gene expression analyses and to support research on PFAS mode of action and nephrotoxicity in human kidney epithelium.
