Self-mating in Leishmania major was demonstrated by crossing two clonal lines of L. major Friedlin in their natural sand fly vector. We used the hybrid progeny to study the contribution of sexual reproduction in generating genotypic heterogeneity in Leishmania and compared it to clonal growth in the sand fly and culture by analyzing multiple parental subclones. WGS and bulk RNA-seq revealed changes in heterozygosity and ploidy that are specific to mating.
