Barcode library sequencing from a plasmid amplicon - supplementary files

Sequencing of a 30N barcode plasmid library, which was used to produce lentiviral particles for in utero next-generation single-cell lineage tracing of ectoderm and mesoderm in mouse embryos. This information is used to flag barcodes that are over-represented in a library and could potentially confound clonal analysis. plasmid_lib_R1.fastq.gz is a raw sequencing file of the barcode region PCR amplicon. This file was processed in order to extract 30N barcodes from a specific location in the amplicon and generate a csv. file extracted_barcodes_plasmid.csv. This file also contains counts of how many times a specific barcode is found in the dataset. No correction was done for the barcode similarity (such as Hamming distance). Scripts for barcode extraction are found at the associated github repository.
