This study employed 16S rDNA and ITS sequencing techniques to investigate the diversity and composition of bacterial and fungal communities in the soil of the sand hills in Poyang Lake. A comparative microbiome analysis will be performed across three chronosequence stages: (1) non-<i> Vitex rotundifolia</i> L. f. areas, (2) 1-year<i> Vitex rotundifolia</i> L. f. growth areas, and (3) multi-year <i>Vitex rotundifolia</i> L. f. growth areas, to assess successional microbial shifts. And will study the relationship between soil physicochemical properties (environmental factors and enzyme activities) related to the nitrogen cycle and microorganisms. The functions of soil microorganisms will be predicted to identify metabolic pathways related to different growth areas of <i>Vitex rotundifolia</i> L. f.. This study aims to provide insights into the ecological mechanisms of growth of<i> Vitex rotundifolia</i> L. f., providing scientific data for standardized cultivation in Poyang Lake's desertified areas, and offering governance approaches for the ecological restoration of deserts in humid regions, thus achieving the goal of ecological conservation (lucid waters and lush mountains) and sustainable socio-economic benefits (golden and silver mountains).
