BRCA1 NGS promoter methylation assay

In order to identify putative markers for platinum sensitivity in a triple negative breast cancer cohort, epigenetic down regulation of the BRCA1 gene was evaluated for 212 primary breast carcinoma samples via next generation sequencing promoter methylation assay. The primary tumour samples were collected as part of the TNT - Triple Negative breast cancer Trial (ISRCTN97330959, NCT00532727, CRUK/07/012). BRCA1 promoter methylation status was established via bi-sulphite sequencing using a 2-step amplicon-based approach. The methylation profiling of the primary tumour samples was performed using an Illumina MiSeq sequencer.
