In this study, we focused on the time-course of transcriptional changes in freshly isolated human PBMCs 4, 8, 24 and 48 h after onset of stimulation with the active vitamin D metabolite 1a,25-dihydroxyvitamin D3 (1,25(OH)2D3) compared to vehicle (0.1% EtOH). Overall design: PBMCs of a healthy male donor were isolated within one hour after peripheral blood collection and exposed for 4, 8, 24 or 48 h to either 10 nM 1a,25-dihydroxyvitamin D3 or solvent (0.1% EtOH). All experiments were performed in three repeats. Total RNA was extracted and subjected to RNA-seq analysis.
