We designed several ZIKV vaccines. Single vaccination conferred sterilizing immunity against ZIKV. To study the mechanism of these vaccines, we did a single cell sequencing experiment to characterize the repertoire of antibodies induced by these ZIKV vaccines.Single-cell suspensions were obtained from the lymph nodes of immunized mice. Sample processing for single B cell receptor (BCR) V(D)J clonotype was done using Chromium Single Cell 5′ Library and the Gel Bead Kit following the manufacturer’s user guide (10x Genomics, Pleasanton, CA, CG000086_SingleCellVDJReagentKitsUserGuide_R- evB).Single-cell BCR V(D)J Libraries were prepared following the manufacturer’s user guide, and profiled using the Bioanalyzer High Sensitivity DNA kit (Agilent Technologies, Santa Clara, CA) and quantified with kapa Library Quantification Kit (Kapa Biosystems, Wilmington, MA). Libraries were sequenced by paired-end sequencing (2 × 150 bp) on an Illumina NovaSeq (Illumina, San Diego, CA). BCL data were converted to demultiplexed FASTQ files using Illumina bcl2fastq 2.20.
