This pilot study developed a system-specific approach to capture fish communities in kelp forests, by analysing fine-scale spatio-temporal patterns in eDNA signal, as well as testing the effect of biological replication and pooling of replicates on species detection. We targeted the hypervariable region of the 12S rRNA gene (163-185bp) using the Mifish-U-F/R primers (Miya et al., 2015).
