Glioma cells must respond to the adverse tumor microenvironment via regulating the translation of specific mRNAs. In a CRISPR screen, we identified that ribosome quality control via sensing ribosome stalling and collisions by EDF1 and ZAK is essential to protect glioma cells against ferroptosis induction. Thus, we aimed to further elucidate their role in glioma pathophysiology. We generated knockout (KO) cell lines for either ZAK or EDF1 using U87 malignant glioma cells. We exposed the cells to Erastin or RSL3 to induce ferroptosis followed by RNA-seq analysis.
