Assessment of genomewide nascent transcription activity and transcriptomic profiles across different stages of directed differentiation from human pluripotent stem cells (hPSC) into small intesitnal organaoids Overall design: Global nascent transcirption activity was determined by chromatin run-on sequencing (ChRO-seq) and transcriptomic profile was determined by bulk RNA-seq. The stages of hPSC directed differentiation included human embryonic stem cell (hESC), definitive endoderm (DE), intestinal spheroids representing duodenal region (Duo), intesitnal spheroids representing ileal region (Ile), Duo intestinal organoid, and Ile intesitnal organoid.
