EAMO-BBMO time-series: rDNA 18S V4 ASV table (dada2)

This repository contains the 18S rRNA gene ASV table generated from rDNA metabarcoding of samples collected at two coastal microbial time-series observatories located at contrasting latitudes: the Equatorial Atlantic Microbial Observatory (EAMO, 6°S) and the Blanes Bay Microbial Observatory (BBMO, 41°N, Mediterranean Sea). Monthly surface seawater samples were collected over 41 months (April 2013–August 2016) and size-fractionated into small (0.22–3 µm) and large (>3 µm) fractions. DNA was extracted and PCR amplified targeting the V4 region of the 18S rRNA gene (Stoeck et al., 2010), followed by Illumina sequencing. The ASVs were generated and taxonomically assigned using the DADA2 pipeline (Callahan et al., 2016) with the Protist Ribosomal Reference database (PR2, version 4.14, (Guillou et al., 2013)⁠). Samples with suboptimal sequencing were removed.The raw DNA sequences obtained in this study were deposited in the European Nucleotide Archive (http://www.ebi.ac.uk/ena) under accession number PRJEB48035 for the BBMO, and in the NCBI (https://www.ncbi.nlm.nih.gov/) under accession number PRJNA414763 for the EAMO.
