E14.5-born neurons at P3 repress L4 neuron-type and induce L2/3 neuron-type genes upon Kir2.1 in utero electroporation

Experimental in vivo hyperpolarization of E14.5 apical progenitors shifted their transcriptional programs with precocious generation of intermediate progenitors, and a forward shift in the laminar, molecular, morphological and circuit features of their neuronal progeny. In particular, we reported an overall repression of L4-type neuronal genes and a premature induction of L2/3-type neuronal genes following Kir2.1 overexpression. We performed bulk RNA sequencing of P3 neurons electroporated at E14.5 with mCherry-reporter plasmid or Kir2.1-GFP plasmid, and at E15.5 with mCherry-reporter plasmid.
