We sequenced the transcriptome of young (6-8 week) and middle-aged (8-10 month old) C57BL/6 female mice that were naïve or had injured ventrolateral spinal cord white matter. We found several differentially expressed genes, many suggesting an altered immune response to injury with advancing age. Naïve or injured (72 hours after the local injection of lysolecithin) ventrolateral white matter of the spinal cord was isolated using laser capture microdissection. RNA was then extracted using RNeasy MiniKit (Qiagen) as per manufacturer’s instruction and sequenced using the Illumina NextSeq500. Tissue was pooled from 5 animals per condition and 3 replicates were sequenced for the analysis.
