3 different MOLM14 cell line xenografts (CLDXs) on NSG mice, treated with venetoclax plus AraC. MOLM-14 cells were injected into mice for a Cell Line Derived Xenograft (CLDX). Briefly, mice (6-9 weeks old) were sublethally treated with 20 mg/kg of busulfan, 24 hours later AML cells were washed in PBS, and suspended in Hank’s Balanced Salt Solution (HBSS) at a final concentration of 2x106 cells per 200μL of HBSS per mouse for tail vein injection. All experiments include male and female mice and experimental groups were randomly assigned. 10 days later, 1 week of treatment begin, consisting in 5 days of AraC treatment (30 mg/kg) by intra-peritoneal injection, in association with 7 days of venetoclax treatment (100 mg/kg) by oral gavage. After one week of treatment, mice bone marrow and spleen were collected. Then viable bone marrow human AML cells (CD45+CD33+AnnexinV-) were sorted and RNA-seq was processed. The experiment was done 3 times into 3 different MOLM14 CLDX.
