Effect of long-term exercise training on lncRNAs expression in the vascular injury of insulin resistance

The present study identified a panel of dysregulated lncRNAs and mRNAs that might serve as potential biomarkers relevant to the vascular injury in insulin resistance induced by high-fat diet. Exercise might alleviate the vascular injury in insulin resistance via targeting FR402720 and FR030200. In this study, 24 male C57BL/6J mice were randomly divided into three groups: control group (ND, n=8), high-fat diet group (HF, n=8), and high-fat diet plus exercise training group (HE, n=8). Glucose tolerance test, insulin tolerance test, fasting plasma glucose, serum insulin level, insulin resistance index, and morphological observations with the electron microscope on the aorta endothelium were applied after 12-week intervention to evaluate the effects of exercise on insulin sensitivity and vascular injury in the three groups. An lncRNA microarray analysis was applied to investigate the global differential expressions of lncRNA and mRNA in aorta endothelium among three groups and the results were further verified by qPCR. Hypergeometric distribution analysis was further applied to figure out the possible functions of these aberrantly expressed lncRNAs.
