This study used dual-RNAseq to compare the transcriptional profiles of mice fibroblasts infected with the JJ21 strain, linked to oral transmission, and the MG strain, from chronic infection. JJ21 exhibited up-regulation of immune evasion and stress response genes, reflecting adaptations to oral transmission. At 24 hours post-infection, host cells showed elevated immune recognition and pro-inflammatory responses, shifting to stress and antioxidant activity by 72 hours. JJ21 also showed higher expression of virulence factors like trans-sialidases and GP63, as well as differential regulation of mucins and dyneins compared to MG. These findings provide insights into strain-specific adaptations and immune responses, informing strategies for managing Chagas disease.
