Wild type HEK293 parental cells and 2 wild type clones and three ADARZαmut (Za domain mutation:N173A/Y177A) HEK293 clones were stimulated for 16 hours with 1000 U/mL IFN-α. Total RNA was purified using RNeasy columns (QIAGEN) with on-column DNase I digestion. RNA integrity was tested with the Agilent RNA 6000 Pico Kit (Agilent; 50671513). Sequencing libraries were prepared using the Stranded mRNA ligation kit (Illumina) with the Ribo-Zero Gold kit (Illumina) for ribosomal depletion. Deep sequencing of the libraries was performed on an Illumina Novaseq6000 generating 150 bp paired-end sequencing reads.
