This study was designed to investigate the toxicological effects of MEHP on TM3 Leydig and TM4 Sertoli cells. For experiments, TM3 Leydig and TM4 Sertoli cells were seeded at 1.5 × 105 cells/ml in 2 ml/well in 6-well plates. After 24 hours, cells were exposed to 100 uM mono-(2-ethylhexyl) ester (MEHP; AccuStandard, ALR-138N), a major metabolite of DEHP in vivo, for another 72 hours. MEHP was dissolved in dimethyl sulfoxide (DMSO; Sigma-Aldrich, D2438), with 0.1% DMSO treatment used as a control.
