This study focuses on the genome sequencing and assembly of Pleurotus ostreatus strain PC80, aiming to provide a high-quality genomic resource. The genome was assembled using PacBio HiFi, Illumina, and Hi-C sequencing technologies, resulting in a genome of approximately 40.7 Mb distributed across 15 scaffolds, 10 of which exhibit telomere-to-telomere structures. The N50 of the assembly is 3.6 Mb. Hi-C technology was used to anchor the genome onto 13 pseudo-chromosomes, demonstrating high completeness and accuracy (98.8% BUSCO score). These results provide a valuable resource for genetic, molecular biological research, and applications in P. ostreatus. The availability of this genome not only enhances the research potential of P. ostreatus in fields such as food science, medicine, and environmental management, but also lays a solid foundation for future genomic research and molecular breeding.
