MicroRNAs (miRNAs) are small non-coding RNA molecules that regulate gene expression by either translational inhibition or mRNA degradation. However, the role of miRNAs in inflammation induced by flagellin (ligand of TLR5) has not been fully determined, Here, we identified differentially expressed miRNAs in murine bone marrow-derived dendritic cells (BMDCs) between flagellin (FliC) treatment and medium alone using miRNA microarray. And we further evaluated the role of miRNAs in inflammation induced by flagellin. Murine BMDCs were generated from C3H/HeJ murine bone marrow in vitro cultured in presence of rmGM-CSF and rmIL-4. The CD11c+ BMDCs were sorted using anti-CD11c-coated magnetic beads with the auto-MACS system. The purified BMDCs were stimulated with 100 ng/mL flagellin (FliC) or medium for 24 h. The differentially expressed miRNAs between flagellin-stimulated group and mock group were screened using miRNA microarray. Three flagellin-stimulated samples and two mock samples were included in the experiment.
