An integrated DNA barcode dataset of carrion- and dung-associated Diptera from the Abruzzo, Lazio and Molise National Park

This dataset accompanies the data paper “An integrated DNA barcode dataset of carrion- and dung-associated Diptera from the Abruzzo, Lazio and Molise National Park” (Biodiversity Data Journal, in review). Flies were collected with baited traps (decaying fish, tiger feces and rhinoceros feces) at six sites (three forest and three pasture sites) located in Zone A, Zone B and outside the Abruzzo, Lazio and Molise National Park (central Apennines, Italy), in two sampling sessions between 27 April and 19 May 2023. A 418 bp fragment of the mitochondrial COI gene was sequenced with Oxford Nanopore MinION for 2,011 specimens, which were clustered into 211 Molecular Operational Taxonomic Units (MOTUs; objective clustering at 3% in INTEGRATAX) belonging to 31 Diptera families. Representative vouchers of each MOTU were identified morphologically and photographed with an Entomoscope photomicroscope. Voucher specimens are deposited in the Museum of Zoology, Sapienza University of Rome (MZUR). Files: PNALM_Diptera_occurrences_DwC.csv – occurrence records of all 2,011 specimens in Darwin Core format. PNALM_Diptera_barcode_data.xlsx – specimen-level data (collection metadata, MOTU membership, morphological and barcode-based identification), list of MOTUs, and haplotype richness and nucleotide diversity per MOTU. The README sheet describes all columns. PNALM_Diptera_barcode_data_Specimen_data.csv, PNALM_Diptera_barcode_data_MOTU_list.csv, PNALM_Diptera_barcode_data_Haplotype_diversity.csv – CSV versions of the three data tables of the Excel file. PNALM_Diptera_COI_sequences.fasta – COI barcode sequences of all 2,011 specimens. Voucher imaging_PNALM.zip – focus-stacked images of one representative voucher for 210 of the 211 MOTUs (the voucher of MOTU 99 was lost).
