New compounds from the platinum metallodrug family have shown very promising results in the fight against cancer, thanks to the introduction of a specific ligand (the NHC carbene ligand) that brings exceptional stability and reactivity to these systems. Although numerous chemical studies have led to a better understanding of the reactivity of these systems, their stability in a cellular environment, the valency and the stereochemistry around the metal center remain unclear. Our objectives are to identify the exact nature of the NHC platinum complexes in the cell and compare the data with those obtained in solution. The key will be to confirm that the NHC carbene ligand remains coordinated on the metallic center, which will confirm the difference in reactivity with so-called classical anticancer platinum compounds, such as cisplatin. Overall, these studies will provide a better understanding of the metabolism of these complexes in cancer cells and their mechanism of action.
