Mapping-by-sequencing of the sus2 mutation

To pinpoint the mutation responsible for the sus2 locus, we conducted mapping-by-sequencing on an F2 population originating from a cross between a sus2 mutant and a wild tomato plant, Solanum pimpinellifolium (accession LA1589). By analyzing genome-wide allele frequencies in two pools, one comprising 19 randomly chosen mutant plants and the other 25 randomly chosen wild-type plants, we were able to locate the genomic region that contains the mutation causing the sus2 phenotype.
