RNA-seq analysis of Near-Isogenic Lines (NILs) in Secale cereale

This dataset comprises RNA sequencing of young leaves of tall and dwarf Near-Isogenic Lines (NILs) at the shooting stage, with five biological replicates per genotype. Total RNA was extracted using the RNeasy Plant Mini Kit (QIAGEN) following the manufacturer's protocol, and RNA quality and integrity were assessed using a Nanodrop spectrophotometer (ThermoFisher Scientific) and Agilent 2100 Bioanalyzer. High-quality RNA samples were used to construct directional RNA-seq libraries with the NEBNext® Ultra™ II Directional RNA Library Prep Kit (NEB) and sequenced on the Illumina NovaSeq 6000 platform in paired-end 150 bp mode. This dataset supports a multidisciplinary study investigating the genetic and molecular basis of dwarfism conferred by the dwarfism causing gene in NILs, integrating phenotypic evaluations, gibberellin sensitivity assays, linkage mapping, and transcriptomic profiling. The data will facilitate further exploration of regulatory pathways and candidate genes influencing plant height.
