<b>CD276 Emerges as a Promising Diagnostic and Prognostic Biomarker for Bladder Cancer through Bioinformatics and Clinical Research</b>

Objective: To determine the expression of CD276 in bladder cancer, and explore the relationship between CD276 expression and clinicopathological characteristics and prognosis.Methods: The RNA sequencing expression profile and clinical information of 436 cases of bladder cancer were downloaded from the TCGA database in UCSC Xena(https://xena.ucsc.edu/). The expression level of CD276 in bladder cancer and para-cancer tissues was retrospectively compared and analyzed. The relationship between CD276 expression and clinical stage, pathological grade, survival prognosis (PFS, OS, DSS) and other related factors was analyzed by R language. The clinical data were retrospectively analyzed. 133 bladder cancer tissue samples and para-cancer tissues from the First Affiliated Hospital of Soochow University from January 1, 2016 to September 30, 2022 were selected, and the expression of CD276 at the protein levels in 133 pairs of bladder cancer tissues and para-cancer tissues was analyzed by immunochemistry. The study further assessed the link between CD276 expression within the tumor tissues and clinical characteristics, as well as its impact on survival prognosis.Results: Bioinformatics analysis revealed that CD276 expression was higher in tumor tissues than in para-cancer tissues (p<0.05), and high expression of CD276 predicted poor prognosis. Immunochemistry experiment confirmed that the expression of CD276 was higher in bladder cancer tissues, and patients with higher pathological grades and poor prognosis showed higher levels of CD276.Conclusion: Considerably overexpressed in bladder cancer, CD276 directly associates with higher pathological grades, advanced stages, the likelihood of distant metastasis, and worse prognoses. Hence, CD276 has potential utility as a prognostic molecular marker for bladder cancer.
