Single-cell RNA-sequencing of osteocyte-derived mitochondria recipient cells in the intra-tibial injection of LLC1 cells murine model.

LLC1 cells were injected into the tibia of DMP1-Cre PhAM mice to study the intercellular mitochondrial transfer in the bone microenvironment. We used single-cell RNA sequencing (scRNA-seq) to analyze the cellular heterogeneity of osteocyte-derived mitochondria recipient cells. Overall design: LLC1 cells were injected intra-tibially into DMP1-Cre PhAM mice. 14 days later tumor-bearing tibia were dissected and enzymatically dissociated into single cell suspension. Osteocyte-derived mitochondria recipient cells were isolated by fluorescence-activated cell sorting (FACS) according to the presence of Dendra2 signal and analyzed using scRNAseq.
