In this study, we selectively knocked down CX3CR1 in cochlear macrophages of C57BL/6 mice using an adeno-associated virus (AAV)-based approach. Five months post-injection, mice in the CX3CR1 knockdown group exhibited delayed ARHL progression and improved outer hair cell survival compared to controls. Bulk RNA-sequencing analysis revealed a downregulation of immune-related pathways, notably reduced expression of Cathepsin C (CTSC) and suppression of TNF signaling, in the CX3CR1-deficient cochleae.
