We used single cell RNA sequencing (scRNA-seq) with TotalSeq-A (BioLegend) cell hashing to investigate the immune cell distribution in the decidua and myometrium of pregnant mice at E7.5. C57BL6 (Hashtag A0306) and Il33cit/cit (IL-33-deficient) (Hashtag A0309) dams were mated with C57BL6 studs. At E7.5, live CD45.2+ cells from the decidua and myometrium of pregnant were stained separately with TotalSeq-A (BioLegend) cell hashing antibodies, isolated by fluorescence-activated cell sorting (FACS), pooled at 1:1 ratio, and analyzed by scRNA-seq.
