To gain insight into the molecular mechanism underlying HuR regulation of ependymal cell differentiation and cilia formation, we performed RNA-immunoprecipitation followed by high-throughput sequencing (RIP-seq) in primary culture ependymal cells from neonatal mouse brain.Next,mice primary culture ependymal cells were treated with actinomycin D to halt de novo transcription and RNA-seq was performed 0 and 5 hour later to obtain the ratio of mRNA levels for each gene in order to measure their stability.
