The goal of this experiment was to determine whether transcriptionally similar ESC-derived and primary donor cells give rise to similar donor-derived cells following transplantation into bleomycin injured lungs. Adult 129X1/S1 mice were given 1.5U/kg bleomycin intratracheally. 10 days later mice received either 4e6 mCh+ ESC-derived tip-like cells (no lentiviral label) or 4e6 primary tip-like cells (labeled with a DsRed lentivirus). At 59 days post-transplantation the lungs of these mice were digested and sorted using FACS. Donor-derived cells were isolated based on the expression of mCherry or DsRed and endogenous cells were collected from the same lungs. 2 separate mice were pooled for both transplant conditions.
