To elucidate the mutational interplay of Ezh2, Runx1 and Bcor we transplanted mice transplanted with LSKs transduced with sgRNAs in all combinations and observed them for development of myeloid leukemia and subsequent expression analysis of whole bone marrow by RNA-Seq. Overall design: LSK cells expressing SpCas9 were transduced with sgRNA combinations and transplanted into irradiated C57Bl/6J mice. BM was harvested after development of a leukemia or after 6 months and used for RNA extraction.
