Advances in long read sequencing motivates the development of new methods for DNA amplification. The T7 replisome is a multiprotein complex capable of highly processive DNA synthesis, and was used in DNA amplification reactions of lambda phage genomic DNA. Nanopore sequencing uncovered a relationship between polymerase exonuclease activity and template-switching in the amplification reaction, and provided a mechanistic description of the process.
