Evaluating Aquaporin-5 (Aqp5) functions in mouse pyloric cancer stem cells

To study Aqp5 functions in pyloric cancer stem cells, we isolated GFP+ putative stem cells and GFP- tumour epithelial cells from Aqp5 wildtype and Aqp5 knockout pyloric mouse tumours to understand the functions of Aqp5 in these cancer stem cells. Overall design: Aqp5-eGFP-ires-CreERT2; Apcfl/fl; Ptenfl/fl; KrasG12D/+ mice with and without an Aqp5 KO allele were induced with two doses of 4mg tamoxifen and the resultant tumours harvested after 10 weeks, with 3 mice analysed per condition. FACS was performed to isolate GFP+ and GFP- tumour epithelial cell populations from each sample for RNAseq.
