Rivael v0.2.0: Blood-Brain Barrier Crossing Drug Candidates and Clinical Trial Gaps for DIPG/Diffuse Midline Glioma

Analysis of 1,097 biomedical papers identified 13 drug candidates that cross the blood-brain barrier and target pathways dysregulated in DIPG (Diffuse Intrinsic Pontine Glioma) but have never been tested in this disease. Three candidates — inavolisib (PI3K inhibitor), avutometinib (RAF/MEK inhibitor), and SCH772984 (ERK inhibitor) — have zero published papers connecting them to any form of pediatric glioma. Four additional candidates (cobimetinib, alpelisib, MDP5, mirdametinib) have fewer than 3 papers each. Six adult-to-pediatric translation gaps were identified where drugs studied in adult glioblastoma have never been tested in pediatric DIPG. DIPG has zero FDA-approved therapies and median survival of 9-11 months in children. All findings represent untested computational hypotheses requiring preclinical validation. The dataset includes plausibility assessments, PubMed novelty verification across 5 search variants per drug, suggested preclinical experiments, and pediatric-specific safety analyses.
