Overexpression of the oncogene HPV8 in keratinocytes of mice causes spontaneous papillomas to form in the skin. To study the expression profiles of skin cancer-associated fibroblasts, we performed RNA-sequencing of fibroblasts FACS-sorted from back skin tumors of HPV8 overexpressing mice. Overall design: CD45-negative/CD140a-positive live cells (fibroblasts) were FACS-sorted from back skin papillomas/tumors (Tu) of HPV8 overexpressing mice and the intact back skin (BS) of their age-matched littermates. Two tumors of 70- and 78-week-old animas were analyzed, along with six BS samples of 68-, 69- and 70-week-old animals, which were pooled to achieve adequate fibroblast cell numbers. The expression profiles of the sorted fibroblasts were analyzed by RNA-sequencing.
