将基因编码的GSH/GSSG (氧化性谷胱甘肽与还原型谷胱甘肽比值)荧光蛋白传感元件和亚细胞结构靶向序列“植入”急性髓系白血病细胞(AML),进而构筑了可稳定表达GSH/GSSG传感元件的活细胞传感器;根据线粒体膜间隙标志性蛋白凋亡诱导因子(AIF)在受到刺激后位移出线粒体的过程,设计并构建了一种荧光蛋白传感器,用来表征线粒体内外膜的损伤。

We integrated genetically encoded fluorescent protein-based sensing elements for GSH/GSSG (ratio of reduced glutathione to oxidized glutathione) and subcellular targeting sequences into acute myeloid leukemia (AML) cells, thereby constructing live-cell biosensors that stably express the GSH/GSSG sensing elements. Based on the translocation of apoptosis-inducing factor (AIF, a marker protein of the mitochondrial intermembrane space) out of mitochondria upon stimulation, we designed and constructed another fluorescent protein-based biosensor to characterize the damage of mitochondrial inner and outer membranes.
