AMT2 and AMT5 were 6mA de novo methyltransferases of Tetrahymena thermophila. Knockout of AMT2 and/or AMT5 abolished methylation on a large majority of methylatable ApT genomic positions. The marked decrease in 6mA levels resulted in a significant reduction in the survival rates of sexual progenies, underscoring the importance of de novo 6mA in progeny development. To clarify the effects of AMT2/5 knockout on progeny, we employed RNA-seq.
