The shell of <i>Nephelium lappaceum</i> (rambutan), an agro-industrial byproduct, is a rich source of bioactive phenolic acids and anthocyanins with potential applications in food and pharmaceutical industries. However, their direct use is limited by instability and low bioavailability. <b>Methods:</b> Phenolic-rich extracts were obtained from rambutan shell and microencapsulated using maltodextrin by spray drying. The chemical composition was determined by HPLC, while FTIR, SEM, and XRD were applied for structural and morphological characterization. Bioactivities were evaluated through antioxidant, antimicrobial, antibiofilm, antitumoral, and hemolytic assays. <b>Results:</b> The extract exhibited high phenolic content, with protocatechuic, syringic, and m-coumaric acids as major constituents. Encapsulation efficiency was 84.86 ± 0.26%, and FTIR, SEM, and XRD confirmed successful entrapment in amorphous, smooth microcapsules. Strong antioxidant activity was recorded (DPPH IC₅₀ = 19.86 µg/mL), together with selective antibacterial effects against <i>Pseudomonas aeruginosa</i> and <i>Burkholderia cepacia</i> (MBC = 62 mg/mL). Significant inhibition of biofilm formation was observed in <i>Staphylococcus aureus</i> and <i>Listeria monocytogenes</i> (&gt;80% inhibition at 1 mg/mL), whereas no antifungal activity was detected against <i>Candida</i> spp. Moreover, the extract reduced viability of tumor cells while sparing normal fibroblasts, highlighting selective antitumoral effects. Hemolysis remained below 2% at all concentrations, confirming biocompatibility. <b>Conclusions:</b> Rambutan shell microcapsules represent a stable, safe, and multifunctional system, combining potent antioxidant, selective antimicrobial, antibiofilm, and antitumoral properties with excellent hemocompatibility. These findings support the valorization of rambutan shell as a sustainable source of bioactive compounds, contributing to the United Nations Sustainable Development Goals (SDG 12: Responsible Consumption and Production; SDG 3: Good Health and Well-Being). <b>Keywords: </b>Phenolic acids, Spray-dried microcapsules, Agro-industrial byproducts, Bioactive compounds, Nutraceutical applications<br>
