We carried out RNA-seq analysis to identify genes that are potentially regulated by PBX4 in hindlimb bud cells of the E11.5 mice by comparing gene expression in KO vs WT mice. Overall design: Hindlimb buds were extracted from E11.5 embryos using forceps under a stereoscope. Total RNAs were isolated using TRIzol (15596-026; Invitrogen). High-throughput sequencing libraries were constructed using the NEB-Next Ultra RNA Library Prep Kit for Illumina.
