Genomic characterization of murine TP53-mutant acute myeloid leukemia

In this study we generated a novel mouse model to study the pathogenesis of therapy-related myeloid neoplasms from TP53-mutant clonal hematopoiesis. Mice carrying inducible Trp53 missense mutations (in mono- or biallelic configuration) were subjected to sublethal gamma-irradiation to mimic genotoxic therapy. After development of malignancies, acute myeloid leukemia cells were sorted into pure populations and subjected to whole genome sequencing to characterized their genomic aberrations.
