Pulmonary hypertension (PH) is a chronic vascular disease characterised, amongst other abnormalities, by hyperproliferative smooth muscle cells and a perturbed cellular redox and metabolic balance. Oxidants induce cell cycle arrest to halt proliferation; however, little is known about the redox-regulated effector proteins that mediate these processes. Here, we report a novel kinase-inhibitory disulfide bond in cyclin D-CDK4 and investigate its role in cell proliferation and PH. We used an Affymetrix microarray to look at the gene expression profile of mouse lungs exposed to normobaric hypoxia (10% oxygen) or normoxia and identified genes that were differentially expressed in response to hypoxia. C57BL/6J mice were subjected to either normoxia (21% oxygen) or normobaric hypoxia (10% oxygen) for 3 days and then anaesthetized with an intraperitoneal injection of pentobarbitone (300 mg/kg) supplemented with heparin (150 U). mRNA extraction from the lung was performed using the RNeasy Plus Universal kit (Qiagen). ~5 μg of SPIA-amplified complementary DNA was fragmented and biotin-labelled using the Encore Biotin module (NuGEN), and hybridisation cocktails were prepared as recommended by NuGEN. Samples were hybridized to Mouse Gene 2.0 ST arrays using standard protocols as recommended by the manufacturer (Affymetrix).
