The transcriptomes of single cells from human bone marrow were investigated at single cell level, with the aim to reconstruct the development of human dendritic cell lineages. 399 human bone marrow mononuclear cells were isolated into 96 well plates via fluorescence-activated cell sorting (FACS). CD34+ cells from the following phenotypic spaces were sequnced at single cell level: CD38-CD45RA-hematopoietic stem cells (HSC) and multipotent progenitors (MPP), CD38+CD45RA- common myeloid progenitors (CMP), CD38-CD45RA+ lymphoid-primed multipotent progenitors (LMPP) and CD38+CD45RA+ granulocyte-macrophage progenitors (GMP).
